Moderna’s 100% Stock Jump Isn’t Just Hype — mRNA Just Won Its First Big Cancer Battle

Most people wrote off Moderna after the COVID vaccine boom faded. They saw it as a one-hit wonder that rode a pandemic to fortune, with no meaningful follow-up. This week’s Phase 3 win for its personalized melanoma mRNA vaccine changes that narrative completely. It’s not just one more incremental drug trial success. It’s proof that the mRNA platform can deliver on decades of promises for treating cancer. That’s why investors boosted Moderna’s market cap by more than 100% in a single day of trading.
Moderna and Merck’s vaccine, called intismeran autogene, hit both primary endpoints in the Phase 3 trial. The trial included 1,137 patients with high-risk melanoma, all of whom had tumors removed surgically before enrollment. One group got the vaccine plus Merck’s immunotherapy Keytruda. The other group got Keytruda alone. Earlier data from the combination showed a 49% reduction in the risk of recurrence or death, and a 59% reduction in distant metastasis or death, compared to Keytruda alone. Results are confirmed to be statistically significant and clinically meaningful, with full data coming later at a medical conference.
For patients with advanced stage 3 or 4 melanoma, current five-year recurrence-free survival ranges from 20 to 35%. Industry clinicians say this new combination could potentially double that survival rate. The vaccine is built from a sample of the patient’s own tumor. It trains the immune system to recognize unique mutations specific to that patient’s cancer. Side effects are minimal compared to traditional chemotherapy. 80% of adverse reactions in earlier small trials resolved on their own without additional intervention.
Investors reacted immediately to the news, with Moderna’s stock more than doubling in early trading. Merck’s shares also climbed, leaving Merck with a $371 billion valuation and Moderna at roughly $62 billion. Many clinicians and analysts argue Moderna remains incredibly undervalued even after the jump. The platform’s value extends far beyond this single melanoma vaccine. This success paves the way for mRNA to target dozens of other cancer types in coming years. It also vindicates the accelerated development path created by Operation Warp Speed for COVID vaccines.
Most coverage ignores a key underlying driver of demand for this treatment. Melanoma rates are rising globally, and climate change is making the problem worse. Higher UV radiation and hotter temperatures increase risk, while sunscreen use continues to drop across populations. It is a major global health issue, with Australia already facing extraordinarily high prevalence. Rates are growing across all ethnic groups, not just Caucasians. Most oncologists are expected to embrace the treatment quickly, so approval is unlikely to face major hurdles.
Moderna will be acquired by a large pharma player within the next 18 months as the platform’s value becomes impossible to ignore.
